The method of isolating Pseudomonas aeruginosa from the root tissues of apple trees was referenced from Lv Nana [18]. After culturing at 28°C for 2 days, bacteria that emitted fluorescence under ultraviolet light were selected and purified for preservation. Following the method of Ji Wenxiu et al. [19], the isolated strains were enriched for ACC deaminase activity. After enrichment, 100μL of bacterial suspension was taken and spread onto ADF solid medium, and then cultured at 25°C for 3 days. After multiple purifications, the strains were preserved on TSB solid medium and stored in a refrigerator at 4°C for later use.
