CBM-Based ELISA for Polysaccharide Quantification: A Novel Approach Using Carbohydrate-Binding Modules
Quantification is crucial for the research and development of polysaccharides. Currently, achieving a simple and specific quantification of polysaccharides remains a challenging task. The use of antibodies in enzyme-linked immunosorbent assay (ELISA) has been proven to be a simple and specific quantification strategy. However, acquiring antibodies for polysaccharides is a complex process. On the other hand, the carbohydrate-binding module (CBM) demonstrates the ability to specifically recognize and bind carbohydrates, and can be obtained efficiently. In this study, we verified the feasibility of a CBM-based ELISA for the quantification of polysaccharides by replacing antibodies with CBMs. As a case study, we utilized hyaluronic acid (HA).
