Immunosuppressive Effects of PTC-0432253 and PTC-0432366 on T Cell Proliferation and IFN-gamma Expression

This study investigated the effects of PTC-0432253 and PTC-0432366 on Treg suppression, finding that both compounds significantly suppressed T cell proliferation in a dose-dependent manner and increased IFN-gamma expression within Foxp3+ cells, suggesting potential immunosuppressive activity. \n\nThe nTreg suppression assay was performed at four ratios (nTreg:T=1:1/0.5:1/0.25:1/0.125:1). From the Treg expansion results, PTC-0432253 and PTC-0432366 showed no effects on the expression of Foxp3 compared with the DMSO group. Both PTC-0432253 and PTC-0432366 could increase the expression of IFN-gamma within Foxp3+ cells. \n\nAdditionally, PTC-0432253 and PTC-0432366 also demonstrated a dose-dependent suppression of T cell proliferation. At a ratio of 1:1, both compounds significantly suppressed T cell proliferation compared to the DMSO group. As the ratio decreased to 0.5:1, 0.25:1, and 0.125:1, the suppressive effects of the compounds on T cell proliferation became even more pronounced. \n\nIn summary, PTC-0432253 and PTC-0432366 showed no effects on Foxp3 expression but increased IFN-gamma expression within Foxp3+ cells. Additionally, both compounds demonstrated a dose-dependent suppression of T cell proliferation. These results suggest that PTC-0432253 and PTC-0432366 have potential immunosuppressive effects and could be further investigated as potential therapeutic agents.
