This study investigated the effects of PTC-0432253 and PTC-0432366 on Treg-mediated suppression of CD4+ and CD8+ T cell proliferation and IFN-gamma release. From the Treg suppression results, nTregs could suppress CD4+T and CD8+ T cell proliferation in the DMSO group. For CD4+ T proliferation, PTC-0432253 promoted the suppression at all ratios tested. PTC-0432366 could only rescue the suppresion at 1:1 but promoted the suppression at 0.5:1/0.25:1/0.125:1. For CD8+ T proliferation, PTC-0432253 promoted the suppression at 1:1/0.5:1/0.25:1. PTC-0432366 could only rescue the suppresion at 1:1 but have no effects on the suppression at 0.5:1/0.25:1/0.125:1. For the ELISA results, the release of IFN-gamma decreased with the increasing number of Tregs in DMSO group which is coordinated with the proliferation results. And PTC-0432253 and PTC-0432366 have no effects on T cells without Tregs. Both PTC-0432253 and PTC-0432366 could promote the release of IFN-gamma at all ratios tested. This suggests that both PTC-0432253 and PTC-0432366 can enhance the production of IFN-gamma when Tregs are present. However, it is important to note that these compounds do not have any effect on T cells in the absence of Tregs.
