nTreg Suppression Assay Results: PTC-0432253 and PTC-0432366 Effects on Treg Expansion and Suppression

Attached please check the results and raw data of the nTreg suppression assay. 4 ratios were performed in this round (nTreg:T=1:1/0.5:1/0.25:1/0.125:1). From the Treg expansion results, PTC-0432253 and PTC-0432366 showed no effects on the percentage of Foxp3 compared with the DMSO group. Both PTC-0432253 and PTC-0432366 could increase the expression of IFN-gamma within Foxp3+ cells.

From the Treg suppression results, nTregs could suppress CD4+T and CD8+ T cell proliferation in the DMSO group. For CD4+ T proliferation, PTC-0432253 promoted the suppression at all ratios tested. PTC-0432366 could only rescue the suppression at 1:1 but promoted the suppression at 0.5:1/0.25:1/0.125:1. For CD8+ T proliferation, PTC-0432253 promoted the suppression at 1:1/0.5:1/0.25:1. PTC-0432366 could only rescue the suppression at 1:1 but have no effects on the suppression at 0.5:1/0.25:1/0.125:1. For the ELISA results, the release of IFN-gamma decreased with the increasing number of Tregs in the DMSO group. And PTC-0432253 and PTC-0432366 have no effects on T cells. Both PTC-0432253 and PTC-0432366 could promote the release of IFN-gamma at all ratios tested.
