Polysaccharide Concentration Determination using Enzyme Solution and DMMD
About 2g freeze-dried sample was mixed with enzyme solution, distilled water (5ml), and heated at 60°C for 1 hour. After adding 2.5ml DMMD solution and agitation, absorbance at 500nm was measured using a cuvette to determine polysaccharide concentration. The enzyme solution contained 50mmol/L KH2PO4 buffer (pH=5.0), 150g/ml papain, 5mmol/L cysteine, and 5mmol/L EDTA.
