EGCG-Modified OVA Induces Tolerogenic DCs and Suppresses Allergic Asthma in AIT
EGCG covalently modified OVA induces tolerogenic DCs and suppresses OVA-induced allergic asthma in AIT.
Allergy immunotherapy (AIT) is a promising strategy for treating allergic diseases. One approach involves inducing tolerance by exposing patients to allergens in a controlled manner. However, conventional AIT often suffers from low efficacy and a risk of adverse reactions. Therefore, developing novel AIT strategies with enhanced efficacy and safety is crucial.
Epigallocatechin gallate (EGCG), a major polyphenol found in green tea, has been shown to possess immunomodulatory properties. Our previous studies have demonstrated that EGCG can induce tolerogenic dendritic cells (DCs), which are essential for establishing immune tolerance.
In this study, we investigated the therapeutic potential of EGCG covalently modified ovalbumin (OVA) in AIT. We found that EGCG-OVA induced tolerogenic DCs that suppressed OVA-induced allergic asthma in a mouse model. Mechanistically, EGCG-OVA-induced DCs exhibited enhanced expression of tolerogenic markers, such as CTLA-4 and IL-10, while reducing the expression of pro-inflammatory cytokines, such as IL-12 and TNF-α. Furthermore, EGCG-OVA-induced DCs effectively suppressed the activation and differentiation of OVA-specific T cells, leading to reduced airway inflammation and hyperresponsiveness in OVA-challenged mice.
Our findings suggest that EGCG-modified OVA holds promise for developing novel AIT strategies. This approach effectively induces tolerogenic DCs, leading to the suppression of allergic asthma in a preclinical model. Further studies are warranted to evaluate the efficacy and safety of EGCG-modified OVA in humans.
