本课题旨在解析缺血心肌微环境和移植MSCs能量代谢特点。本课题拟开展以下研究内容:1. 结合蛋白质和代谢组学方法,分析小动物(小鼠心肌梗死后缺血心肌微环境中三大代谢物质葡萄糖、脂肪酸、支链氨基酸的信号通路—底物利用—产物堆积的网络变化;2. 制定缺血心肌微环境中三大代谢物质代谢特点和分子机制的参考标准;3. 建立MSCs移植后在体多模态示踪技术,完成移植后的MSCs 生物行为学规律的在体示踪;4. 解析归巢到心肌缺血微环境中的MSCs葡萄糖、脂肪酸、支链氨基酸的信号通路—底物利用—产物堆积的网络变化;5. 制定归巢到缺血心肌微环境中的MSCs能量代谢研究参考标准。
The aim of this project is to analyze the energy metabolism characteristics of ischemic myocardial microenvironment and transplanted MSCs. The following research contents will be conducted: 1. Using proteomics and metabolomics methods, analyze the network changes of signal pathways, substrate utilization, and product accumulation of the three major metabolites glucose, fatty acids, and branched-chain amino acids in the ischemic myocardial microenvironment after myocardial infarction in small animals (mice); 2. Develop reference standards for the metabolic characteristics and molecular mechanisms of the three major metabolites in the ischemic myocardial microenvironment; 3. Establish in vivo multimodal tracing technology for the biological behavior of transplanted MSCs after transplantation; 4. Analyze the network changes of signal pathways, substrate utilization, and product accumulation of glucose, fatty acids, and branched-chain amino acids in MSCs homing to the ischemic myocardial microenvironment; 5. Develop reference standards for the energy metabolism of MSCs homing to the ischemic myocardial microenvironment.
