This review aims to explore the role of autophagy in regulating stem cells and promoting oral tissue regeneration. We discuss the mechanisms by which autophagy helps oral stem cells cope with survival stress and provide an overview of the autophagy machinery in eukaryotic cells. We also examine how autophagy contributes to the different components of oral tissue regeneration.

Furthermore, we delve into the molecular mechanisms that underlie autophagy-regulated oral tissue regeneration and how it can be modulated by various approaches such as small molecule drugs, biomaterials, exosomes/RNAs or other specific treatments. Lastly, we discuss new perspectives on autophagy manipulation and its potential applications in oral tissue regeneration.

Despite the progress made in this field, the exact mechanism by which autophagy contributes to oral tissue regeneration remains unclear. Further mechanistic studies are necessary to fully understand the therapeutic potential of autophagy in tissue regeneration. We hope that this review provides new insights into human tissue regeneration and stimulates further research on autophagy in oral tissue and other regenerative processes.
