A range of computational combinatorial approaches were integrated to expedite the improvement of D-lactonohydrolase activity whilst minimizing costs. These strategies offer a versatile framework for enhancing the performance of enzymes with unclear catalytic mechanisms and lacking experimental structures, thereby facilitating their application in synthetic biology.
