Polysaccharide quantification is crucial for the research and development of polysaccharides (Noyes et al., 2014). It has recently gained widespread attention due to the research boom in functional products based on polysaccharides. There is an urgent need for polysaccharide quantification to ensure quality control in the pharmaceuticals, functional food, and cosmetics industries (Nishinari & Takahashi, 2003). However, the chemical heterogeneity and diversity of polysaccharides present a significant challenge, making their simple and specific quantification extremely difficult, thus hindering the development and application of polysaccharides (Li et al., 2022).

Currently, colorimetric methods such as the phenol-sulfuric acid assay (Dubois et al., 1951), anthrone-sulfuric acid assay (ROE, 1955), carbazole-sulfuric acid assay (Bitter & Muir, 1962), dische method (Bitter & Muir, 1962), and methylene blue method (Soedjak, 1994) are commonly used for polysaccharide quantification. Among them, the phenol-sulfuric acid assay remains the most popular method. These methods are widely employed due to their simplicity, but they suffer from poor accuracy and low specificity. Moreover, large instruments such as high-performance liquid chromatography (HPLC) (Galant et al., 2015), gas chromatography (GC) (Lv et al., 2015), and mass spectrometry (MS) (Šimek et al., 2020) offer alternative platforms for polysaccharide quantification. However, these tools are limited by their laborious processes and expensive equipment. Additionally, enzymatic methods are highly effective for quantifying specific polysaccharides by utilizing the catalytic abilities of enzymes. Although enzymatic methods have been applied to the quantitative analysis of glucans (Mccleary & codd, 1991), fructans (McCleary et al., 2000), and starch (McCleary et al., 1997), they suffer from incomplete transformation of polysaccharides and the difficulty of obtaining specific polysaccharidases. Therefore, there is a continued need for advanced research to develop simple and specific methods for polysaccharide quantification
