miR-137-5p inhibits USP30 expression by directly targeting its 3'UTR
A dual luciferase reporter gene assay (Figure 3E) was conducted to investigate the interaction between miR-137-5p and the 3' untranslated region (3'UTR) of USP30. Results showed that the luciferase activity of the wild-type (WT) USP30 3'UTR was significantly decreased upon exposure to the miR-137-5p mimic. Conversely, no significant change in luciferase activity was observed in the mutant type (MUT) of the ZC3H12B 3'UTR (p > 0.05). These findings suggest that miR-137-5p directly targets the 3'UTR of USP30, leading to the inhibition of its expression.
