Emerging evidence suggests that TGFβ has suppressive effects on angiogenesis. However, our findings indicate that TGFβ's anti-angiogenesis role is weakened in the tumor context, which is crucial for promoting vigorous tumor angiogenesis. Through in vivo models, we observed that TGFβ could suppress physiological angiogenesis, such as zebrafish vessel formation. However, it was unable to suppress tumor angiogenesis. Both in vitro and in vivo studies revealed that hypoxia plays a critical role in switching TGFβ's function, allowing tumors to overcome anti-angiogenesis control. This may explain why tumor angiogenesis remains highly active even in TGFβ-enriched microenvironments
