The Pommier research group demonstrated [24] that Top1mt is a direct mitochondrial target of Lam-D. Mitochondria contain their own DNA (mtDNA), and the only specific mitochondrial topoisomerase in vertebrates is topoisomerase I (Top1mt). In vitro, Lam-D captures Top1mt and induces Top1mt cleavage complexes (Top1mtcc). Using single-molecule analysis, the authors show that Lam-D slows down Top1mt supercoiling relaxation and strongly inhibits Top1mt re-joining compared to the ineffective action of camptothecin on Top1mt. In living cells, we found that Lam-D rapidly accumulates in mitochondria, induces Top1mtcc, and causes mtDNA damage.
