The research team led by Pommier demonstrated [24] that Top1mt is a direct mitochondrial target of Lam-D. Mitochondria contain their own DNA (mtDNA), and the only specific mitochondrial topoisomerase in vertebrates is topoisomerase I (Top1mt). Lam-D captures Top1mt in vitro and induces Top1mt cleavage complexes (Top1mtcc). Using single-molecule analysis, the authors showed that compared to the ineffective action of camptothecin on Top1mt, Lam-D slows down Top1mt supercoil relaxation and strongly inhibits Top1mt re-aggregation.
