This study successfully constructed a eukaryotic expression vector, pEGFP-N1-MIC1, designed to overexpress the mouse macrophage inhibitory cytokine-1 (MIC-1) gene. The vector's efficacy was confirmed through enzyme digestion, DNA sequencing, and transfection into the MFC cell line. Transfected cells demonstrated a significant increase in MIC1 protein expression levels. Furthermore, pEGFP-N1-MIC1 transfection reduced the migration and colony formation capabilities of MFC cells, suggesting its potential role in inhibiting tumor progression. This research contributes valuable insights into MIC-1's functional role and establishes pEGFP-N1-MIC1 as a powerful tool for future investigations into MIC-1's therapeutic potential, particularly in the context of gastric cancer.
