In this study, an unidentified G-specific alginate lyase gene sequence was excavated from the CAZy database and heterologously expressed in E. coli. The purified recombinant enzyme was analyzed for its enzymatic and kinetic properties. Additionally, UPSEC-DAD/MS and HPAEC-PAD/MS techniques were used to determine the degradation mode of substrate cutting site, subsite specificity, and cutting mode of the enzyme. These findings provide a clear degradation pathway for G-specific alginate lyase, which is important for studying the structure-activity relationship of alginate and preparing active oligosaccharides.
