In summary, this study identified that Pdi1, a member of the protein disulfide isomerase family, is essential for the formation of the hydrophobic layer of Aspergillus conidia. Pdi1 is involved in the correct folding and assembly of RodA, the main component of the hydrophobic layer. The absence of Pdi1 results in the destruction of the hydrophobic layer and loss of conidia hydrophobicity. The study also found that the retention of any trx active domain of Pdi1, N-terminal transmembrane signal, and C-terminal endoplasmic reticulum retention signal is necessary for maintaining the normal hydrophobicity of conidia. These findings provide a new perspective for the prevention and control of mold pollution and infection, by targeting the regulatory elements involved in the formation of the hydrophobic layer.
