DNA methylation is an important epigenetic modification that plays a crucial role in gene expression regulation, embryonic development, cell differentiation, tumorigenesis, and other biological processes. Abnormal changes in DNA methylation are closely associated with the occurrence and development of various diseases. Therefore, accurate and efficient detection of DNA methylation status is of great significance for the diagnosis and treatment of diseases. With the advancement of technology, various DNA methylation detection methods have been developed, including methylation-specific PCR, methylation-sensitive restriction enzyme digestion, methylation-sensitive sequencing, methylation-sensitive PCR, and methylation-sensitive high-throughput sequencing. These methods have different advantages and disadvantages, and can be selected according to the research objectives and experimental conditions. This review provides an overview of the recent research progress in DNA methylation detection methods, offering important references for researchers
