PEG-Based Crystallization Screen for Biological Macromolecules
PEG-Based Crystallization Screen for Biological Macromolecules
This screen includes a wide range of conditions using polyethylene glycol (PEG) of different molecular weights and other common precipitants. It is designed for screening the crystallization of biological macromolecules, such as proteins, by varying the pH value, buffer system, and additives.
A. PEG Solutions with Different Molecular Weights:
- A11: 0.1 M Imidazole pH 7.0, 25% v/v Polyethylene glycol monomethyl ether 550* B2: 0.1 M MES monohydrate pH 6.0, 20% v/v Jeffamineᆴ M-600ᆴ pH 7.0* B5: 0.1 M Sodium citrate tribasic dihydrate pH 5.5, 22% w/v Polyethylene glycol 1,000* B8: 0.1 M BIS-TRIS pH 6.5, 20% w/v Polyethylene glycol 1,500* B11: 0.1 M MES monohydrate pH 6.0, 20% w/v Polyethylene glycol monomethyl ether 2,000* C5: 0.1 M Sodium citrate tribasic dihydrate pH 5.5, 18% w/v Polyethylene glycol 3,350* D1: 0.1 M Sodium citrate tribasic dihydrate pH 5.0, 10% w/v Polyethylene glycol 6,000* D3: 0.1 M BIS-TRIS propane pH 9.0, 30% w/v Polyethylene glycol 6,000* D8: 0.1 M BIS-TRIS pH 6.5, 16% w/v Polyethylene glycol 10,000
B. PEG Solutions with Additional Precipitants and Additives:
- E11: 1.8 M Ammonium sulfate, 0.1 M BIS-TRIS pH 6.5, 2% v/v Polyethylene glycol monomethyl ether 550* G1: 0.10% w/v n-Octyl-b-D-glucoside, 0.1 M Sodium citrate tribasic dihydrate pH 5.5, 22% w/v Polyethylene glycol 3,350* G8: 0.2 M Sodium formate, 0.1 M BICINE pH 8.5, 20% w/v Polyethylene glycol monomethyl ether 5,000* H4: 15% v/v 2-Propanol, 0.1 M Sodium citrate tribasic dihydrate pH 5.0, 10% w/v Polyethylene glycol 10,000
Note: This is just a selection of conditions, and a full crystallization screen may include a wider range of PEGs, buffers, salts, and additives.
