Transcriptome analysis of tobacco at different time points after treatment with tobacco mosaic virus (TMV) was performed by transcriptome sequencing analysis on samples collected at 0 h, 3 h, 6 h, 12 h, 24 h, and 48 h after TMV inoculation. Compared to 0 h after inoculation, 7191, 5207, 1944, 7768, and 4026 differentially expressed genes were identified at 3 h, 6 h, 12 h, 24 h, and 48 h after inoculation, respectively. GO functional enrichment analysis showed that these differentially expressed genes were enriched in response to stimuli, defense response, pathogenesis, peroxidase activity, hormone activity, immune response, autophagy, and other functions. KEGG pathway enrichment analysis revealed that differentially expressed genes were enriched in biological processes related to plant-pathogen interactions, plant hormone signal transduction, peroxisomes, regulation of autophagy, and biosynthesis of cutin, suberin, and wax. These results indicate that the differentially expressed genes respond to the stress of TMV through these biological processes and signal transduction pathways. Further analysis revealed that 27 members of the tobacco NtTCP gene family were differentially expressed, and the expression levels of NtTCP5, NtTCP7, NtTCP13, NtTCP18, NtTCP20, NtTCP29, NtTCP36, and NtTCP38 genes were quantitatively validated at different time points after TMV infection, and the results were consistent with the transcriptome data, indicating that the expression of some NtTCP genes was affected by TMV infection and may be related to the response of tobacco to TMV infection.
