Heart failure is a complex condition that represents the final outcome of most cardiovascular diseases, yet the molecular mechanisms underlying this debilitating condition remain largely elusive. This study aimed to systematically investigate the underlying molecular mechanisms of heart failure, along with potential diagnostic and therapeutic targets, using bioinformatics tools. To this end, we obtained a total of 16 samples, including 8 from healthy individuals and 8 from heart failure patients, from publicly available datasets (GSE8331 and GSE76701). After removing any batch effects, we performed a differential analysis and identified 185 differentially expressed genes. Our enrichment analysis revealed that the molecular mechanisms of heart failure were predominantly associated with pathways related to immune function, inflammation, and metabolism.

We further investigated the immune cell infiltration patterns in heart failure samples and found that Tgd cells and Neurons were significantly enriched, whereas pDCs and NKTs were more abundant in healthy tissues. To identify the most important genes involved in heart failure, we performed a PPI network analysis and identified EGR1, EGR2, FOS, and FOSB as hub genes. These genes have been previously implicated in various physiological and pathological processes, and our findings suggest their potential involvement in the development and progression of heart failure.

Overall, our study sheds light on the complex molecular mechanisms underlying heart failure and provides important insights into potential diagnostic and therapeutic targets. Further studies are warranted to validate our findings and explore the clinical implications of these findings.
