I hope this email finds you well. Attached, please find the results and raw data of the nTreg suppression assay. In this round, we performed four ratios of nTreg:T (1:1, 0.5:1, 0.25:1, 0.125:1).

From the Treg expansion results, both PTC-0432253 and PTC-0432366 showed no effects on the percentage of Foxp3 compared to the DMSO group. However, both compounds were found to increase the expression of IFN-gamma within Foxp3+ cells.

Regarding the Treg suppression results, nTregs were able to suppress the proliferation of CD4+T and CD8+T cells in the DMSO group. PTC-0432253 promoted suppression at all tested ratios for CD4+T cell proliferation. On the other hand, PTC-0432366 only rescued suppression at a ratio of 1:1 but promoted suppression at ratios of 0.5:1, 0.25:1, and 0.125:1.

For CD8+T cell proliferation, PTC-0432253 promoted suppression at ratios of 1:1, 0.5:1, and 0.25:1. However, PTC-0432366 only rescued suppression at a ratio of 1:1 and had no effects on the suppression at ratios of 0.5:1, 0.25:1, and 0.125:1.

In the ELISA results, the release of IFN-gamma decreased as the number of Tregs increased in the DMSO group. PTC-0432253 and PTC-0432366 had no effects on T cells. However, both compounds were found to promote the release of IFN-gamma at all tested ratios.
